①Reagent preparation area: The main instruments and equipment include refrigerators, ultra-clean tables, samplers, mixers, balances, centrifuges, etc. In addition to dedicated samplers and balances, they should also be calibrated regularly. Reagent sub-packaging should be carried out in an ultra-clean bench. The amplification reaction mixture should be of molecular biology grade. After the reagents are prepared, there should be quality inspection: firstly whether there is contamination, and secondly, detecting the amplification effect of the reagents.

②Specimen preparation area: The main equipment includes biological safety cabinets, samplers, centrifuges, incubators, mixers, etc.

③Amplification area: The main instruments are nucleic acid amplifier, ultra-clean table, micro-sampler, centrifuge, and movable ultraviolet lamp. The thermal cycler needs to be calibrated. And equipped with UPS power supply to prevent the impact of power outage on the amplification effect due to voltage fluctuations.

④Analysis area: The main instruments and equipment include microplate reader, plate washer, sampler and water bath, etc. The area of the buffer room in the clean experimental area cannot exceed one-eighth of the main laboratory, which is stipulated.

Main structure: The main body is color steel plate and aluminum alloy profile. All internal corners and external corners in the room are made of aluminum alloy 50 internal rounded aluminum, which solves the problems of easy pollution, dust accumulation, and difficult cleaning. The structure is firm, the lines are concise, the appearance is generous, and the airtightness is good.

Standard four-zone separation and air pressure adjustment: The PCR process is divided into four independent experimental zones for reagent preparation, specimen preparation, PCR amplification, and product analysis. There is an overall buffer corridor throughout the area. Each independent experimental area is equipped with a buffer zone, and each area is adjusted by air pressure to prevent the reagents and specimens from aerosol contamination during the entire PCR experiment and reduce the pollution of the amplified products to personnel and the environment.

The exhaust fans of buffer Ⅰ, buffer Ⅱ and PCR amplification zone can be opened to exhaust air. The external wall and each door of the experimental area are equipped with return air vents with adjustable air volume, and the air is ventilated into the room through the return air vents.

Disinfection: UV lamps are installed on the top of the three experimental areas and the three buffer zones and inside the transmission window for disinfection. In the reagent preparation area and the specimen preparation area, mobile ultraviolet lamps are also installed to locally disinfect the experimental table.

Stainless steel transfer window: reagents and specimens are transferred through mechanical interlocking stainless steel (electronic interlocking is not recommended) to ensure that reagents and specimens are not contaminated during the transfer process (personal shunt).

Ground: It is recommended to use PVC coil ground or artesian ground for the ground, which has good integrity. Easy to clean and corrosion resistant.

Lighting: Purifying lamps should be selected for lamps, which can achieve the characteristics of easy cleaning and no dust accumulation.
