The instruments and equipment in the reagent preparation area mainly include refrigerator, super clean table, sample dispenser, mixer, balance and centrifuge. In addition to being dedicated, the sample dispenser and balance should also be calibrated regularly. The reagent dispensing shall be carried out in the ultra clean platform. The mixed solution of amplification reaction shall be of molecular biology grade. After the preparation of the reagent, there shall be quality inspection: first, check whether there is pollution; second, check the amplification effect of the reagent.
The instruments and equipment in the specimen preparation area mainly include biosafety cabinet, sampler, centrifuge, incubator, mixer, etc
Amplification area: the main instruments in the amplification area are nucleic acid amplification instrument, ultra clean table, micro sampler, centrifuge and mobile ultraviolet lamp. The amplifier needs to be calibrated. UPS power supply is also provided to prevent the impact of power failure on amplification effect due to voltage fluctuation.
Analysis area: the instruments and equipment used in this area include microplate reader, plate washer, sample feeder and water bath box.
It is stipulated that the buffer room area of PCR experimental area shall not exceed one eighth of the main laboratory
1. Main structure:
The main body is color steel plate and aluminum alloy profile. All internal and external corners in the room are made of aluminum alloy 50 fillet aluminum, so as to solve the problems of easy pollution, dust accumulation and difficult cleaning. The structure is firm, the lines are simple, beautiful and generous, and the sealing property is good.
2. Standard four zone separation and air pressure regulation:
The PCR process was divided into four independent experimental areas: reagent preparation, sample preparation, PCR amplification and product analysis. There is an overall buffer corridor in the whole area. Each independent experimental zone is provided with a buffer zone. Meanwhile, each zone is adjusted by air pressure to prevent reagents and samples from being polluted by aerosols during the whole PCR experiment and reduce the pollution of amplification products to people and the environment.
The exhaust fans of buffer zone I, buffer zone II and PCR amplification zone can be turned on to exhaust outside. Air return vents with adjustable air volume are installed on the outer walls and doors of the Experimental Zone, and the air is ventilated indoors through the air return vents.
Ultraviolet lamps are installed at the top of three experimental areas and three buffer areas and inside the transfer window for disinfection.
Mobile ultraviolet lamps are also set in the reagent preparation area and specimen preparation area to locally disinfect the experimental table.
4. Stainless steel transfer window:
Reagents and samples are delivered through mechanically linked stainless steel (electronic linkage is not recommended) delivery windows to ensure that reagents and samples are not contaminated during delivery (Figure shunt).
It is recommended to use PVC coiled material floor or self leveling floor for good integrity. Easy to clean and corrosion resistant.
Lamps and lanterns shall be clean lamps and lanterns, which shall be easy to clean and free of dust.
In the process of construction, attention should be paid to:
● standardized installation process and comprehensive service process.
● strictly follow the installation process of scientific design.
● before installation, the following installation conditions need to be determined, such as: power supply voltage, power incoming line position, telephone network line incoming line position, water inlet pressure, minimum installation space, ground flatness, positions of vent holes, water inlet pipes and downpipes, etc., and ideal space size.
